Gubenzhike Recipe on Respiratory Mucosa Immune Mechanism of COPD Model Mice FREE TO VIEW

PURPOSE: To observe the effects of Gubenzhike recipe on pulmonary function, sIgA level in respiratory tract, epithelial gamma delta-T cell in lung and other indices express of COPD mice. Explore the effect of Gubenzhike recipe on immune mechanism of COPD model mice in the respiratory mucosa.

METHODS: The mouse COPD model was established by exposure to cigarette smoke in combination with intra-nostrils injection of lipopolysaccharide. The mice were divided into three groups: negative control group, model group and Gubenzhike recipe group; Use the methods of HE eosin staining and electronic microscope to observe the morphology characteristic of COPD model; Test the pulmonary function by animal pulmonary function machine; Use the ELISA to test the sIgA level in respiratory, the KGF level in supernatant of lung homogenate, the IFN-γ and IL-4 level in blood serum and the IL-17 level in supernatant of epithelial lymphocyte culture; Use the method of Flow cytometry to test the expression of epithelial γδ T cell in lung.

RESULTS: The morphology characteristic is up to COPD; The FEV0.1, FEV0.2, FEF25-75, PEF and the content of sIgA in the upper respiratory tract in Gubenzhike recipe group had significant difference compared with those in model group (P<0.05). The negative control group had no significant difference compared with Gubenzhike recipe group. The proportion of γδT cells of model group was higher than negative control group (P<0.05); the proportion of γδT cells of Gubenzhike recipe group was lower (P<0.05); The IL-17 level in supernatant of epithelial lymphocyte culture coincided with the changing trend of γδT cell. The mice serum level of IFN-γ decreased and IL-4 increased significantly in Gubenzhike recipe group than that in c model group (P<0.05) The expression of KGF in lung tissue is decreased in model group (P<0.05). The expression of KGF in lung tissue is higher in Gubenzhike recipe group than those in model group (P<0.05).

CONCLUSIONS: Gubenzhike recipe might improve respiratory mucosal immune function by regulating the biological activity of γδT cells, which reduce the secretion of IL-17; Correcting Thl/Th2 unbalance; Reduce the sIgA level in respiratory tract and promoting the secretion of KGF. So Gubenzhike recipe might not only relieve airway inflammation but also could improve the pulmonary function and respiratory tract mucosa immunity of COPD model.

CLINICAL IMPLICATIONS: It is conducive to the prevention and treatment of COPD.
