PURPOSE: To further investigate the adverse role of LncRNA in the NSCLC.

METHODS: We screened the expression of LncRNA in two NSCLC lines, 95D and 95C with high and low metastatic potential respectively, using the high throughput LncRNA chip and identified high expression of TATDN1, one of lncRNAs, in the 95D cells comparing with low expression of TATDN1-1 in 95C cells. Then RNA interference (RNAi) was used to block TATDN1expression in 95D cells and the cell proliferation, adhesion, migration and invasion of the TATDN1 depleted 95D cells were investigated.

RESULTS: TATDN1was highly expressed in 95D cells lines and NSCLC tissues. When TATDN1-1 in95D cells was blocked the cell proliferation, adhesion, migration and invasion were significantly inhibited. Xenografts of 95Dcell with blocked TATDN1 expression on nude mice grow slowly and the tumor sizes are smaller.

CONCLUSIONS: These data indicate that theTATDN1expression level associated with 95D cells’ higher potential of invasion and metastasis.

CLINICAL IMPLICATIONS: It may be used as a prognostic factor and new therapy target molecule.
