The Effect of Simvastatin on Paracrinal Cytokines Secreted by TGF-β1-Induced AECs FREE TO VIEW

PURPOSE: As the therapy for Idiopathic pulmonary fibrosis (IPF) is limited, which has high incidence and poor prognosis in senior patient, A study was conducted to investigate the effect of simvastatin on paracrinal cytokines secreted by TGF-β1 -induced AECs in vitro, aiming to provide experience for further studys on threapy for IPF.

METHODS: We compared the level of the paracrinal cytokines of ACEs induced by TGF-β1 or not, then compared the same cytokines with or without the Intervention of Sim, including CTGF, PDGF, ET-1 and IGF-1. The levels of those paracrinal cytokines within cells were tested with Western blot (WB) and the levels of cytokines in the incubating culture were tested with ELISA.

RESULTS: 1. Induction of A549 cells by TGF-β1: Comparing with control group, both levels of expression of CTGF, PDGF, ET-1 and IGF-1 within cells and incubating culture are increased (p<0.05).; 2. Inhibition of induced paracrine in A549 cells by Sim: Comparing with TGF-β1 group, both levels of expression of CTGF, PDGF, ET-1 and IGF-1 within cells and incubating culture are decreased after the intervention of Sim (p<0.05).

CONCLUSIONS: 1. TGF-β1-induced A549 cells overexpress CTGF, PDGF, ET-1 and IGF-1. 2. Sim could reverse the overpression of CTGF, PDGF, ET-1 and IGF-1 in TGF-β1-induced A549 cells.

CLINICAL IMPLICATIONS: Our study may provide experience for further studys on threapy for IPF
