PURPOSE: To investigate the Effect of lupus recipe on gene expression of CD134, CD134L and RANTES from lung tissues of lupus –prone BXSB mice.
METHODS: Eighteen male lupus-prone BXSB model mice were divided into three groups: un-treated group; lupus recipe (LR) treated group; prednisone treated group. Meanwhile, 6 syngeneic normal C57BL/6 male non-lupus-prone mice were used as the control group. All model mice were killed in 10 weeks. RT-FQ-PCR was used to detect the mRNA expression of CD134, CD134L and RANTES in lung tissues.
RESULTS: (1) The mRNA expressions of CD134, CD134L and RANTES from lung tissues in un-treated group were obviously higher than those from control groups (p<0.05). The LR or prednisone significantly reduced the CD134, CD134L and RANTES mRNA expressions in LR treated group or prednisone treated group comparing with model group (p<0.05), while the difference between the treated group and the normal group was not significant. (2) There was a positive correlation between CD134L mRNA expression and RANTES mRNA expression in model lung tissues (r =0.793, P<0.05).
CONCLUSION: The abnormal expression of CD134, CD134L mRNA was well evidenced in the lung of lupus-prone BXSB mice.
CLINICAL IMPLICATIONS: Strong expression of RANTES mRNA was likely mediated by the activation of CD134/CD134L signal pathway. LR and corticosteroids could obviously inhibit the mRNA expressions of CD134, CD134L and RANTES.
